We have synthesized a 582,970-base pair Mycoplasma genitalium genome. This synthetic genome, named M. genitalium JCVI-1.0, contains all the genes of wild-type M. genitalium G37 except MG408, which was disrupted by an antibiotic marker to block pathogenicity and to allow for selection. To identify the genome as synthetic, we inserted "watermarks" at intergenic sites known to tolerate transposon insertions. Overlapping "cassettes" of 5 to 7 kilobases (kb), assembled from chemically synthesized oligonucleotides, were joined by in vitro recombination to produce intermediate (...) assemblies of approximately 24 kb, 72 kb ("1/8 genome"), and 144 kb ("1/4 genome"), which were all cloned as bacterial artificial chromosomes in Escherichia coli. Most of these intermediate clones were sequenced, and clones of all four 1/4 genomes with the correct sequence were identified. The complete synthetic genome was assembled by transformation-associated recombination cloning in the yeast Saccharomyces cerevisiae, then isolated and sequenced. A clone with the correct sequence was identified. The methods described here will be generally useful for constructing large DNA molecules from chemically synthesized pieces and also from combinations of natural and synthetic DNA segments. 10.1126/science.1151721. Smith's three-volume theology of culture, the author expands and deepens the analysis of cultural liturgies and Christian worship he developed in his well-received Desiring the Kingdom. He helps us understand and appreciate the bodily basis of habit formation and how liturgical formation--both "secular" and Christian--affects our fundamental orientation to the (...) world. Worship "works" by leveraging our bodies to transform our imagination, and it does this through stories we understand on a register that is closer to body than mind. This has critical implications for how we think about Christian formation. Professors and students will welcome this work as will pastors, worship leaders, and Christian educators. Michael Hornsby-Smith offers an overview of Catholic social thought particularly in recent decades. While drawing on official teaching such as papal encyclicals and the pastoral letters of bishops' conferences, he takes seriously the need for dialogue with secular thought. The 2006 book is organized in four stages. Part I outlines the variety of domestic and international injustices and seeks to offer a social analysis of the causes of these injustices. Part II offers a theological reflection on the characteristics of (...) the kingdom of God which Christians are urged to seek. Part III reviews Catholic social thought in six main areas: human rights, the family and bioethical issues, economic life, social exclusion, authentic development, and war and peace. This article explores how ministerial and leadership formation could be enabled to adopt transformational diaconia in addressing poverty in South Africa, engaging in ways in which pastoral care and leadership formation can respond to the addressing of poverty. The fact that transformation aims at changing the worldviews, paradigms and approaches to life and problem solving informs the author’s concept of transformational diaconia, which was proposed as an aspect of spiritual leadership capital (SLC), defined as, ‘The inner virtues afforded individuals by (...) their spirituality in formulating their leadership paradigms which contributes to social capital formation for addressing social problems’. Spiritual leadership capital is hereby argued to be a transformative spirituality that can enable an understanding and sustainable responses to poverty and other social problems. This is needed for Africa and particularly for the present day South Africa, seemingly a country with the best infrastructure in Africa; yet its poverty seems pronounced because the dregs of apartheid still lurk in the social fibre, where poor people blame rich people for their plight and vice versa. Bowers Du Toit’s view that ‘[m]ost congregations respond to poverty by providing relief and not empowerment’, re-echoes here. From a mixed-methods research, SLC is a theory recently advanced as a congregational development paradigm and a theology of poverty, which views public theology as an educative praxis that can respond to transformational needs in poverty-related contexts. The authors suggest that for a Church that is responsive to the plight of society, fresh empowerment approaches to address poverty are needed. (shrink)

The control of various introduced species brings to the fore questions around how species are categorised as ‘native’ or ‘invasive’, belonging or not belonging. In far north Queensland, Australia, the Cape York region is a complex mixture of land tenures, including pastoral leases, National Parks and Aboriginal land, and overlapping management agreements. Weed control comprises much of the work that land managers in Cape York do. However, different land managers target different introduced species for control, and the ways in which (...) certain species are understood as more or less problematic indicate how land managers understand and seek to order landscapes. Through investigating the various positions that introduced species occupy, I will explore how Cape York emerges as a ‘hybrid landscape’ that is produced in contested, overlapping and ambiguous ways, and is rife with ‘feral dynamics’. In this article the focus is on ritual theory and its relevance for pastoral care during the grief process. For these purposes the first task at hand is finding an appropriate description of what ritual implies, especially in the context of pastoral care. It includes studying different descriptions from different study fields to provide a broad theoretical view of ritual and to identify relevant perspectives. This view is narrowed to ritual as performance as well as legitimisation of experience. Concerning the (...) pastoral aspect of grief care the research mainly focuses on the work of the Dutch practical theologian, Corja Menken-Bekius and the American practical theologian David Hogue. Menken-Bekius' works on ritual from a clinical pastoral perspective while Hogue gives a neuropsychological perspective. Included in the processing of these two views are the works of anthropologist, Roy Rappaport and psychologist, Onno Van der Hart. Finding a description of ritual is not an easy task. The one offered is created with careful consideration of the inclusions and exclusions within the 'idea' of ritual. Meaning is subjective when it comes to ritual. A general meaning might be ascribed to a specific ritual, yet each individual might have a different experience and might attach different meanings. This happens unwittingly and unwillingly. What is found to be important in the rituals proposed in this article, is the ability of rituals to provide a framework of order within a situation of chaos and that it also creates a moment of remembrance. This chapter offers a summary of Empson’s transformation of the traditional literary concept of the genre of pastoral. This involves acknowledging his indebtedness to that traditional concept, but it also involves bringing out the way his redefinition of it as ‘putting the complex into the simple’ importantly depends on an essay by James Smith which brings out a deep internal connection between the traditional category and a range of metaphysical issues, originating in Plato’s concern with the relation between the (...) one and the many, but ramifying in a number of significant ways throughout the subsequent history of philosophy. Considerable controversy was stirred by the contrast between the specific approaches to public policy contained in the first draft of the Catholic bishops' letter on the U.S. economy and the policies favored by the Reagan administration. However, a much more basic contrast actually existed between the bishops' underlying vision of economic life and contemporary capitalism. The pastoral challenges a separation between moral criteria and economic activity that is deeply embedded in modernity itself. Indeed, the splitting off of economic life from (...) its moral-religious matrix is seen by the bishops' critics as a positive, defining feature of democratic capitalism. The critics see the separate economic and moral-religious spheres related by due balance; the bishops, while acknowledging an autonomy to economic life, emphasize that its fundamental choices remain moral. The bishops (and, for different reasons, their critics) have preferred to minimize the contrast between the letter's vision and the contemporary economy. They avoid any clearcut judgment on the economic system by stressing pragmatism and reforms; but implicitly they are granting a strictly conditioned acceptance of reformist capitalism, the condition being the system's openness to questioning and change. Ai° D 20.Como las dos recientes traducciones castellanas del Liber de cau sis tan sólo tienen en cuenta el texto de Pattin, y parecen desconocer las propuestas de Taylon hemos creído conveniente publicar esta tercera traducción, señalando los cambios más importantes respecto a la edición de Pattin siempre y cuando éstos afecten a la traducción. Counterfactual definiteness is supposed to underlie the Bell theorem. An old controversy exists among those who reject the theorem implications by rejecting counterfactual definiteness and those who claim that, since it is a direct consequence of locality, it cannot be independently rejected. We propose a different approach for solving this contentious issue by realizing that counterfactual definiteness is an unnecessary and inconsistent assumption. Counterfactual definiteness is not equivalent to realism or determinism neither it follows from locality. It merely reduces to (...) an incongruent application of counterfactual reasoning. Being incompatible with falsifiability, it constitutes an unjustified assumption that goes against the scientific method rigor. Correct formulations of the Bell theorem’s bases show it is absent either as a fundamental hypothesis or as a consequence of something else. Most importantly, we present a coherent Bell inequality derivation carefully devised to show explicitly and convincingly the absence of incompatible experiments or counterfactual reasoning. Thus, even admitting that counterfactual definiteness could be a consistent assumption, the necessary conclusion is that it is irrelevant for the inequality formulation and can be safely ignored when discussing Bell’s inequality philosophical and physical implications. We present a pragmatic analysis of the different meanings assigned to the term “local realism” in the context of the empirical violations of Bell-type inequalities since its inception in the late 1970s. We point out that most of them are inappropriate and arise from a deeply ingrained prejudice that originated in the celebrated 1935 paper by Einstein-Podolski-Rosen. We highlight the correct connotation that arises once we discard unnecessary metaphysics.

A common mistake present in the derivation of the usually known as the CHSH form of Bell’s inequalities is pointed out. References and comments to the correct approach are given. This error does not alter the final result and only affects the logical consistency of the derivation, but since it seems to be a widespread misconception regarding the roll and interpretation of the of use of hidden variables in Bell’s theorem it is considered to be of general interest.

In a recent article https://doi.org/10.1007/s10699-020-09666-0) Marek Czachor claims that the Bell inequality cannot be proved because variables of complementary measurements cannot be added or multiplied. Even though he has correctly identified the problems existing with the orthodox interpretation of the Bell inequality and dealt with them in an original way, the interpretation he addresses do not pertain to the original formulation given by John Stewart Bell.

A discussion of Nieuwenhuizen’s description for the hidden variables of the detectors in the derivation of Bell’s theorem is presented. This description prevents Bell’s inequalities from being effected. However it will be argued, on mathematical and physical bases, that the flaws attributed by Nieuwenhuizen to Bell’s probability distribution function are unjustified.

We study the coverage of European funds by the mass media and the relationship between media sentiment and public sentiment. We analyze 31,570 media articles published across all European Union (EU) countries between January 1, 2020, and December 31, 2023 — a period significantly shaped by the COVID-19 pandemic and major EU funding initiatives —using the BERTopic modeling technique. We find that public discussions related to European funds can be categorized into 11 distinct topics, ranging from project developments in Central (...) and Eastern Europe to employment and economic measures. Our results show that during the period studied, public sentiment towards European funds is generally stable yet negative, with notable fluctuations corresponding to specific events. Significant positive and negative peaks in sentiment are observed during periods associated with successful project implementations or corruption scandals. We also find significant geographical variation in media coverage and sentiment across EU member states. While Greece, Croatia, and Bulgaria exhibit high media coverage and more negative sentiment, Denmark, Ireland, and Portugal display more positive media sentiments. Our results suggest a negative relationship between media sentiment scores and the proportion of negative responses concerning the EU and the European Parliament (i.e., a proxy for Euroscepticism). Scholars have recently claimed that heritage values are not fixed, but culturally created and historically specific. This article will analyze this statement by looking at how the concept and perception of heritage values themselves have been transformed in the last two centuries. The consequences of these changes will be examined in terms of decisions regarding what constitutes heritage and what should be preserved, the institutions related to new values that have been established and how this has impacted on national legislation (...) and international agreements. It will be argued that there has been a steady growth in the number of heritage value categories taken into account by experts and that this has been accompanied by a displacement in the centrality of some that were previously regarded as the only ones. The main values considered in the article will be historical, aesthetic, natural, anthropological, symbolic, social, and economic. In contrast to the nineteenth and early twentieth centuries, recent decades have seen a shift in focus from the material to the immaterial or intangible aspects of heritage, and from historical to social and economic values. Today, it is difficult to find agreement among professionals on a valid typology of heritage values, as preferences for the sets of categories used for value assessment depend on the country and the experts’ professional background, as well as on the type of heritage and context. The latest shift is linked to a global emphasis on social inclusivity, cultural sustainability and significance. Although this article focuses mainly on Europe, and to a lesser extent on the United States, much of what it is explained can be applied to other areas of the world. (shrink)

Our work aimed to study the relationships between different dimensions of school climate, moral disengagement, empathy, and bullying behaviors (perpetration and victimization). The study sample consisted of 629 students (304 boys and 325 girls) aged 12–14 years (M= 12.55,SD= 0.67). Results showed how different dimensions of school climate predicted moral disengagement, empathy, and victimization, and these, in turn, predicted bullying perpetration. The results show the need to generate favorable educational environments to reduce the levels of moral disengagement and victimization and (...) to increase empathy in students as a strategy to prevent negative consequences related to bullying. La educación superior pasó a desarrollarse en contextos digitales, lo que supuso un rediseño en cuanto a los recursos, instrumentos, modos, estrategias de actuación y medidas a implementar en el proceso de enseñanza-aprendizaje. En este estudio se analiza, desde una mirada cualitativa, las visiones del estudiantado sobre la docencia universitaria en contextos de pandemia. Los resultados evidencian la perspectiva diferencial del alumnado,-entusiasta, crítica, (...) condescendiente y nostálgica- sobre la educacion superior, en atención a su experiencia universitaria, vivida en contextos de profunda excepcionalidad. La Fraternidad de la Flecha Dorada, fundada por la ocultista Maria de Naglowska en el París de 1932, fue una de las distintas órdenes mágicas de la Edad Contemporánea sobre las que se ha investigado en contextos relacionados con el satanismo, aunque en un número muy reducido de obras y nunca con demasiada profundidad. La fraternidad, que destacó por su sacerdocio exclusivamente femenino y el gran protagonismo de la magia sexual, se basaba en la particular e integradora visión teológica de (...) Naglowska, de carácter no dualista, que incluía una trinidad compuesta por Dios, Jesús y, por último, Satán. Sin embargo, ni Satán ni Lucifer ejercen el papel de figura central dentro de este sistema esotérico cuya autora tampoco parece haber ejercido algún tipo de influencia en las bases del fenómeno conocido como «satanismo contemporáneo». Expuesto este fondo, mi comunicación tendrá un objetivo doble: presentar los cimientos teóricos de la Fraternidad de la Flecha Dorada, así como algunos de los rituales destacados de esta orden, para, en segundo lugar, determinar su naturaleza y revisar, debido a las razones mencionadas, la etiqueta de «satanismo» que ha sobrevolado la misma, tratando de clasificarla. Para ello, centraré mi análisis en el marco de las Ciencias de las Religiones, más concretamente en el de los nuevos movimientos religiosos y el esoterismo occidental, abordando los textos de Maria de Naglowska utilizados por la fraternidad. Además, me apoyaré en investigaciones anteriores con el fin de recopilar los datos históricos necesarios para introducir a la orden y a su carismática suma sacerdotisa. (shrink)

Sara Ahmed piensa las emociones, los objetos, los espacios y las orientaciones hacia el mundo a partir de sus efectos. Las emociones son realidades públicas que nos pegan y nos separan de los otros, adquiriendo su significación política en el contacto social. La idea de la pegajosidad y el contacto presupone una subjetividad indisociable de su relación con la alteridad. Para explicar esta relación entre el yo y el mundo, Ahmed hace uso de la fenomenología. Una fenomenología queer consistirá en (...) sacar a la luz el fondo de historias, normas y signos que los espacios normativos y los objetos familiares ocultan al presentarse como atemporales y naturales. En este trabajo propongo que la filosofía de Ahmed permite leer las orientaciones, la subjetividad y las emociones en su dimensión narrativa. El protagonismo que adquieren las historias en la teoría queer de Ahmed es útil para pensar el vínculo entre literatura y política a partir de la recepción y la circulación de las obras. Las narrativas feministas contribuyen a generar extrañamiento respecto del mundo normativo, a dislocar el orden familiar. Estos dislocamientos abren grietas desde las que imaginar, a través de la literatura, maneras no normativas de desear y percibir el mundo. (shrink)

El artículo analiza cualitativamente la relación entre infancia y violencia en el contexto de la guerra civil en El Salvador (1980-1992). La reflexión es vehiculada por el examen de la película Voces inocentes (2004), como una obra socialmente producida y como una representación de la realidad histórica de índole prospectiva. El trabajo destaca el mérito del filme al visibilizar y denunciar la responsabilidad de los sustentadores primarios de la violencia ‒aún hoy impune‒ hacia la infancia salvadoreña, cuyas secuelas psicosociales constituyeron, (...) a la postre, un entorno sociohistórico centroamericano signado por una debacle humanitaria que comenzó hace cuatro décadas, en un tránsito infausto “de la locura a la esperanza”. Para ello, he empleado el marco teórico conceptual sobre la violencia desarrollado por el jesuita vasco-salvadoreño, Ignacio Ellacuría, según el cual, las violencias resultantes solo pueden ser valoradas a partir de la crítica inicial a la violencia originaria de carácter estructural. La Declaración Universal de los Derechos Humanos de 1948 nació y tendió a la expansión del reconocimiento universal de la dignidad humana. Incorporando el pasado, tratando de comprender el sentido de la existencia e incluyendo al mundo en esa búsqueda, aspiró a lo categórico, a conocer la esencia, el valor intrínseco del ser humano, en relación con otros seres. Hoy, en su nombre, dada la actual ruptura con la tradición y la Historia, se ha extendido una nueva y paradójica concepción (...) de lo humano, emancipadora y homogeneizante. Clamando la reivindicación del “yo”, se ha producido una inconsistencia absoluta en la aplicación de la DUDH: no puede seguir llamándose universal una Carta subsumida y aplicada en un contexto radicalmente uniforme. The objective of the article is to problematize the marginal place of the sex trade in the order of mining work, articulated from the psychodynamics of labor policies. We work with critical discourse analysis with individual and group interviews with male miners and with women self-defined as sex workers, from the Antofagasta Region. We conclude that the sex trade and the work of sex workers have a central place in the reproduction of the mining workforce, along with it, the search (...) for psychosexual well-being is satisfied, where sex work enhances the maintenance of the virile image, of a man who is not ready himself to the deconstruction of the hegemonic gender. El aprendizaje de la Historia en Educación Secundaria se relaciona con un proceso memorístico y conceptual que dificulta la formación del pensamiento crítico y reflexivo en el alumnado de esta etapa educativa. Por este motivo, con esta experiencia y desde un enfoque investigativo, hemos fomentado la curiosidad y el interés de los alumnos de 1º de Secundaria por las prácticas religiosas y funerarias que profesaron los romanos. El método de enseñanza-aprendizaje fue activo y participativo, lo que facilitó la indagación y (...) la experimentación del taller “Funus romanorum”. Los resultados mostraron las ventajas de trabajar la arqueología experimental para comprender mejor los hechos acaecidos en la Antigüedad y adquirir contenidos históricos a través de talleres experienciales. In this sense, we seek to explore cases of epistemic stance that indicate epistemological positioning and persuade readers of the veracity of propositions. Our study covers Scottish journal articles published online within 5 days prior to results day. In this article, we are interested in the way the Scottish newspapers deal with the topic of the independence referendum and the degree (...) of commitment in journalists’ choice of linguistic strategies. We analysed the newspaper articles manually but corpus tools were also used in order to verify initial findings and for statistical purposes. The results of the use of epistemic stance devices in the different categories were contrasted and our conclusions showed the preferred legitimising strategies used by journalists in the Scottish newspapers.
